My command I am trying to run is
lilypond -e '#(set-global-staff-size 32)' --pdf myfile.ly

To run successfully your file, i have changed a bit your code and your
command line.
(Don't ask me why your code didn't work)

If you are not in Windows, perhaps you'll have to change "" by '
#(use-modules (guile-user)) gives error messages also for me
