Theoretically, LilyPond understands these characters natively.
However, make sure that you text editor program does (you have to save
your .ly files as UTF-8 encoding).

No, the TeX notation doesn't work, and isn't even needed!
The only situation where you need to type \" is when you want to print
quotes in your text (title, lyrics, etc).

LilyPond is very different from TeX. You do not need to have any tex
distribution installed to work with lilypond. The lilypond examples
are to be simply saved as .ly files on your computer, and then
compiled with lilypond. (you can just double-click on a .ly file to
see the score printed as a PDF file, next to it).

You can also find additional examples on the LilyPond Snippet Repository:

(there is a plugin for LaTeX called lilypond-book, in case you need to
include musical exceprts in a TeX document, but if you only want to
print a score, this will not be useful.)
