The spacing properties require a list, i.e., you need to set one or more of the 
four sub-properties (basic-distance, minimum-distance, padding, 
stretchability). Try

    markup-system-spacing.basic-distance = 20

or

    markup-system-spacing = #’((basic-distance . 20))
