No, it is enabled by default and there is no configuration option for disabling it. There is even a classdef object defined in the core Octave distribution (the InputParser class). What exactly isn't working?
If you simply typed classdef at the octave prompt and got an error, that's because classdef keywords are only recognized inside files. There is no way to define a class at the command line.
